Pectinase Produce The Bacteria Hdym-02 Mutation Breeding

Pectinase is a kind of decomposition of the pectin collectively, contain a variety of components of the composite enzyme. Pectinase widely distributed over higher plants and microorganisms, in certain protozoa and insects are also found. In micro-organisms, bacteria, actinomycetes, yeast and mold can to metabolize and synthesize the pectinase. Pectinase is an important emerging enzymes in the area of industrial production. According to statistics, at present, pectinase accounted for 25 per cent in the world of food enzyme sales.In this paper, we impose HDYM-02 which was isolated from Retting ponds in Heilongjiang Bayan flax Ltd. as the starting strain, we process mutation to use ultraviolet light, and from a large number of mutations to the screening, we select four mutant strains which produce Pectinase and property is stable and Significantly enhance the enzymatic activity: U6, U12, U27, U36, one of U12 when in 6 h the enzymatic activity as the starting strain of 1.44 times. U12 will be as NTG mutation of starting strain, after four cycle of mutation, we screened five mutant strains : N4, N15, N33, N52 . the enzymatic activity of N33 is the 2.2 b times of U12 when in 12h. Finally, making the N33 to UV and NTG composite mutation, we obtain the seven mutants: F6, F18, F22, F35, which the enzymatic activity of F22 at 24 h achieve to 98.33 umol / mL.Finally, we make mutation of the F22 with UV and diethyl sulfate, obtain final mutant F'14 of this research, it's enzymatic activity is 111.27 umol / mL in the 24 h, it is 3.2 times of starting production strains (34.39 umol / mL) .We switch the eventually mutant F'14 to ten generation, and detecting the extinction value of F'14broth with the DNS approach ,it showed that the extinction value of F'14 broth is relatively stable, it illustrate that output of Pectinase is stable and not appear back mutation, and it compare to starting strain the extinction value have greatly improved.
