China's beer production and consumption has been in the first place in the world for six consecutive years. In 2007, our country produced about 35.15 million tons of beer and 600 thousand tons waste beer yeast. Waste beer yeast whose RNA content is 6%~8% is rich in nutrient and it is good as material for RNA production. Using it wisely can create good economic benefits for beer enterprises as well as improve the competitiveness of enterprises after their entering the WTO. It will also help to protect the environment by using waste beer yeast.The experiment in which waste beer yeast was the raw material studied the best extraction technology of salt method, single enzyme method (dispase method andβ-glucanase method respectively) and double enzyme method (dispase method andβ-glucanase method working together) in which the enzyme method is rarely seen in documents. At the same time, the active dry yeast was used as raw material to be compared with the waste beer yeast. Its purpose is to provide facultative raw material for large-scale RNA production.The results showed that the optimized condition using waste beer yeast for RNA extraction with salt method was: temperature 100℃, original pH(5.89~5.95), salt concentration 9, action time 7h, RNA yield 6.28%, purity quotient 91.23%; dispase was better in enzymic method and the best extraction condition was: temperature 55℃, pH6.5, enzyme quantity 3%, action time 1h, RNA yield 3.24%, purity quotient 91.67%. In actual production, the two methods can be selected if the energy and other factors are considered. According to the experimental results, active dry yeast can be used as a raw material supplement for RNA production.
