At present the resistance that pests to Bt desinsection genes has became a hot spots.Therefore native and foreign researchers are trying their best to find a new way to deal with insect resistant .Synthetic Tat protein has the unique feature of entering the cultured cell efficiently in vitro with other protein expression of fusion proteins (Tat-protein) , researchers have introduced Tat peptide into agricultural biological control and also defined synergies Tat to Bt toxin protein Cry1Ac gene and anti-confrontational role in pests grams and the further effect has reached 9.69 times,so construct fusion genes of Bt desinsection genes and Tat protein gene and transform it to mode plants to check its expression in plant system and to definitude the synergism of Tat to Bt desinsection genes in plant system,then break a new using path of Tat and a new way of pests control.The subject increases further reasonable application of Tat in transgenic plants to research the expression of Tat peptide in plants and the further effect to Bt toxin protein Cry1Ac gene. To multiply the Cry1A gene from double transgenic cotton with the method PCR .Reconstruct the Tat codes and design and construct Tat gene sequence, in order to check the expression fusion peptide of Tat in plants ,we use YFP gene sequence as a sign gene to recombinant a complete Cry1A gene vector with Tat gene sequence and YFP gene sequence. In order to research the relations between the genes and the effect of Tat peptide, the subject has constructed pBI121-Cry1A,pBI121-Tat-Cry1A,pBI121-Cry1A-YFP,pBI121-Tat-Cry1A-YFP, and (control)the four fusion gene expression vector. That agrobacterium mediated transformation to tobacco and Arabidopsis, and the establishment of a complete transformation of the model plant system, has made base on further expression of integration of the diamondback moth resistance and virulence of the sensitive stocks, a clear expression of fusion protein in the plant after the adversarial role of pest management theory and practice laid the basis. If the studies further verified Tat in plants can improve efficiency and magical effect of pests grams, and Tat application are of great theoretical and practical significance, and will open a new way to overcome resistance to pests and a new field in Tat application.
