The antler velvet, a famous traditional Chinese medicine, is the non-ossification angle organization of the deer with various kinds of biological activity materials, which can promote organism growth and metabolism, enhance immune function and other special effects. The extraction and application of antler velvet has an important economic value and social benefits.A cationic exchange-HPLC method for determination of sialic acid in antler velvet was newly developed. Researchs on the processes of ultrasonic extraction, supercritical CO2 extraction and homogenate extraction on the antler velvet were conducted.And the optimal extraction condition was determined for the degradation from the antler powder at the same time.HPLC method for determination of sialic acid in antler velvet was presented.The pretreatment of sample and hydrolysis condition were optimized. The chromatographic analytic conditions were as follows:separation column of ZORBAX 300-SCX cationic exchange column, aqueous solution of acetonitrile-0.1% phosphoric acid (90+10, v/v) as mobile phase,1.0 mL/min of flow rate,10.0μL of the injection volume, and the detection wavelength at 205 nm. The sialic acid was good linear correlation in the range of 5-100μg/mL. The detection limit (S/N= 3) was 0.03 g/kg for sialic acid in antler velvet. The recovery was 83.4% with 4.9% of RSD (n=6). The method can be applied to determine the sialic acid content in antler velvet and related products.This paper uses ultrasonic extraction method to extract antler. The effects of temperature, ultrasonic power, ratio of solid to liquid and ethanol concentration were discussed. Under the best extraction condition, the extraction yield can reach to 8.38%.Then the active components was extracted by supercritical CO2.The effects of extraction temperature, pressure, ration of antler and co-solvent, and concentration of co-solvent were investigated. The experimental results show that the extraction yield can reach to 3.86%. Furthermore the extraction of antler velvet by homogenate extraction was primarily studied.To make full use of resources, the alkaline protease Alcalase is employed to degrade the antler powder in the paper. And the optimal extraction condition was determined for the degradation. The results showed that the degradation yield is up to 60.2%. Then the optimal freeze-drying condition of the enzymatic solution is also explored in the study. The best effect of freeze-drying is achieved when the pH of concentrated liquid is 4-5, and the product was made a white powder. The analysis showed that the product containing threonine, histidine, alanine and other 13 kinds of amino acids. The product is expected to become an important nutrient.
