Facing the increasingly serious oil pollution environmental problems, how to utilize the microbes to its many research institutes become free-pollution disposal and academics looked topic. So if we get a plant can effectively degrade petroleum hydrocarbon strains will give the microorganism oil pollution issues to govern provides a new method and means.This experiment through modern biological engineering technology and methods obtained the strains of L5 which can growth by screening take crude oil as the only carbon source and have high petroleum hydrocarbons degradation rate from the oil pollution soil samples and water samples which got from YanChang oilfield. And through the observation on morphological characteristics of strains L5 and carry out a series of physiological and biochemical tests, then according to the《Bergey's Manual of Determinative Bacteriology》and《common bacteria system identification Manual》,with the test results preliminarily determined to identify, can hold coli strains L5 belong to Bacilus spp.. Then we mutagenesist to the strains L5 on the methods of UV mutagenesis,He-Ne mutants and compound mutagenesis, obtained the strains of L(F)52 which can growth by screening take crude oil as the only carbon source and the biggest degradation rate can reach 39.9%,had a significantly improved compared with the departure strains L5. After the genetic stability test proof received L(F)52 strains has the stable ability of growth and stable ability of petroleum hydrocarbon degradation.In order to further understanding and improve the ability of petroleum hydrocarbon degradation of the strains of L(F)52. we take the single factor experiment, make sure that L(F)52' s optimal grow condition:bacteria levitation liquid inoculation quantity 2%, the incubation temperature 30℃, medium pH 7.2 and training at least 12 days, the highest degradation rate can reach 44.5%. Then we analysis each composition of the crude oil which degradated by the strains of L(F)52. The test samples which after degradation process were detected by the production engineering research institute of offshore oil corporation of China and Xi'an petroleum institute, and the results show that after degradation process with contrast oil samples wax, bituminous, gelatin content have significantly reduced. Its means that strains L(F)52 can effective use of the macromolecular oil hydrocarbon.
