High performance liquid chromatography (HPLC) is one of the most important methods for separation and analysis in the subject of chemistry. Almost all the liquid organic, polymer and biological samples can be analyzed by HPLC. It has been widely used in petroleum chemical industry, biochemistry, clinical medicine, foodhyginen, environmental monitoring, medicine industrial inspection and other fields.The combination of HPLC and Chemical derivatization could satisfy the request of the ultra trace analysis of the most materials in nature. Developing new derivatization reagents for HPLC analysis is one of the active research areas for chromatography. In this paper, two novel derivatization reagents were designed and characterized. The spectral properties, optimal derivatization and chromatographic separation conditions were investigated. A pre-column derivatization reversed-phase HPLC method was established and evaluated for the analysis of real samples.Chapter one:The development history and basic theory about high performance liquid chromatography and high performance liquid chromatography coupled with mass spectrometry were simply presented. The theory and application of chemical derivatization with fluorescence derivatization were systematically introduced, cited widely used derivative reagents in recent years.Chapter two:Design and synthesis of two novel fluorescent labeling reagents, 10-ethyl-2-amino-acridone(EAA),2-(4-amino)phenyl-1-hydrogen-Philippine[9,10-d]imidazole(P IA),and followed with their structural and spectroscopic properties.Chapter there:Pre-column derivatization methods for the sensitive determination of fatty acids with 10-ethyl-2-amino-acridone(EAA) as labeling reagents followed by HPLC-MS analysis have been developed. The optimization of derivatization and chromatographic separation conditions were evaluated.Then Put these methods applied to analyze margarine, milk and some pollen samples.Chapter four:Pre-column derivatization methods for the determination of saturated and unsaturated fatty acids with 10-ethyl-2-amino-acridone(EAA) and 2-(4-amino)phenyl-1-hydrog-en-Philippine[9,10-d]imidazole(PIA) as labeling reagents followed by HPLC-MS analysis have been developed. The optimization of derivatization and chromatographic separation conditions were evaluated. The established methods were applied to the determination of different real samples。Chapter Five:Pre-column derivatization methods for the sensitive determination of aliphatic aldehydes and bile acids with 2-(4-amino)phenyl-1-hydrogen-Philippine[9,10-d]imidaz- ole(PIA) and 10-ethyl-2-amino-acridone(EAA) as derivative reagents respectively followed by HPLC-MS analysis has been developed. The optimization of derivatization and chromatographic separation conditions were evaluated. Then, these methods were applied to the determination of real samples, and the results suggested that the established methods have great feasibility.
