Staphylococcus aureus is a bacterium, and is frequently found in the human respiratorytract and on the skin. Although S. aureus is not always pathogenic, it is a common cause ofskin infections, respiratory disease, food poisoning and clinical infection. Its harmfulness islarge, and recent researches have shown that infection caused by S. aureus has an obviousupward trend, so analyzing and detecting S. aureus have the important clinical significanceand use value. S. aureus can be detected by enzyme-linked immune method, but usually usingtraditional methods such as thin layer chromatography and ultraviolet spectrophotometry,however these methods have the low sensitivities and some limitations for measuring S.aureus. Thus the author built a high sensitivity detection method of S. aureus. The theory isthat S. aureus can connect with alkaline phosphatase (AP) according to the specificity of theantigen and antibody, and AP can catalyze disodium phenyl phosphate hydrate (DPPD) for theproduct phenol, DPPD has no impact on B-Z oscillation system, but phenol can disturb theB-Z oscillation system, causing the amplitude and cycle changes of the oscillation system. SoB-Z oscillation system combined with enzyme-linked immune detect the concentrationchange of S. aureus by analyzing the cycle or amplitude change.We designed two kinds of B-Z chemical oscillation system, one was Sulfuric Acid-Acetone-Glucose-Sodium Bromate-Macrocyclic Copper oscillation system, another wasthe Sulfuric Acid-Malic Acid-Sodium Bromate-Macrocyclic Copper oscillation system,then by optimizing the component concentrations of the two systems respectively, we got thebest component concentration, the result for the Acetone-Glucose system was:[H2SO4]=1.05M,[NaBrO3]=0.08M,(Acetone)=0.65M,[[CuL](ClO4)2]=0.05M,(Glucose)=0.06M; the result for the Malic Acid system was:[H2SO4]=1.05M,[NaBrO3]=0.8M,(MalicAcid)=1.5M,[[CuL](ClO4)2]=0.03M. Using the establishing Acetone-Glucose system todetect AP, and analyzing the relationship between the concentration and the amplitudevariation, we found that the correlation coefficient R=0.9984; for testing the phenol, R=0.9921; The Acetone-Glucose system combined with double antibody sandwichenzyme-linked immunoassay detected S. Aureus mycoprotein, the lowest detection limit was1.059710-9g/mL; The Acetone-glucose system combined with Biotin–Avidinenzyme-linked immune detected S. aureus mycoprotein, the lowest detection limit was1.463610-11g/mL. If using the establishing Malic Acid system to detect AP, and analyzing therelationship between the concentration and the amplitude variation, we found that the correlation coefficient R=0.9905; for testing the phenol, R=0.991; The Malic Acid systemcombined with double antibody sandwich enzyme-linked immunoassay detected S. aureusmycoprotein, the lowest detection limit was1.23910-10g/mL; The Malic Acid systemcombined with Biotin–Avidin enzyme-linked immune detected S. aureus mycoprotein, thelowest detection limit was1.35610-11g/mL. Thus the two kinds of B-Z oscillation systemcombined with enzyme-linked immunoassay for detecting S. aureus all have a good linearityand high sensitivity.In this article, B-Z chemical oscillation system’s volume was10mL, the system had agood test repeatability and stability, Malic Acid system and Acetone-Glucose oscillationsystem have a very high correlation for detecting phenol, this provides the basis for a smallcapacity development of the B-Z oscillation system; This detection method, B-Z chemicaloscillation combined with enzyme-linked immunoassay opens up a train of thoughts fordetecting S. aureus, it has the very important practical significance; In addition,enzyme-linked immunoassay combined with B-Z chemical oscillation analysis method canalso be used for detecting other materials’ concentration changes such as drugs,macromolecules, small molecules, etal, This analysis method has a wide application area anda promising prospect, and provides a new detection method for other materials.
