Alkaloids Extracted From Dendrobium And Their Activity To Suppress Caco-2 Cells

Our research took the Dendrobium nobile as the material, used the organic solvent to extract the fat soluble alkaloids and water-soluble alkaloids, purified the water soluble alkaloids and identified their structures. We also investigated the crude extracts and erianin affecting the cell proliferation, apoptosis and cycle, and detected the activity of the cell caspase-3. The Results showed that there were 11.9% alkaloids in the fat-soluble crude extracts, while the alkaloids held 7.93% share in the water-soluble crude extracts. After purified the water-soluble alkaloids by D101 and LH-20, two components named JW-1 and JW-2 were got. The result of 1R identification showed the two components contained similar functional groups, including benzene ring structure, C-O, N-H structure. The erianin and crude extracts all presented dose-and time-dependently inhibited the viability of Caco-2 cells and induced the apoptosis, accompanied by significantly change in cell nuclear morphology and membrane potential. Furthermore, erianin made cell cycle arrest at G2 phase, water-soluble alkaloids made cell cycle arrest at G1 phase. The further study revealed that fat-soluble and water-soluble alkaloids both can induce the caspase-3 activation.The findings in this study have important implications for the development of Dendrobium nobile in anticancer applications, suggesting that crude extracts could exert inhibitory effect on Caco-2 to a degree, and we could consider further research in clinical trials.
