Egg white protein contains essential amino acids and possesses high availability for human. Egg white protein hydrolysates have many bioactivity properties such as antibacterial, antioxidant, antihypertensive. In addition, egg white protein hydrolysate used as natural food antimicrobial agents are widely used in the food industry to inhibit growth of microorganism including foodborn pathogens.Three kinds of egg white from different species (hen, quail, and duck egg) were hydrosylate by using pepsin enzyme at different time (1-5h) and different types of bacteria (S. aureus and E. Coli) to determine the antibacterial activity analysis in the terms of radial difussion assay and flow cytometry. The Hydroxyl radical scavenging ability analyzed by chemiluminenscence system to determine antioxidant activity. Molecular weight of three kinds of egg white protein hydrolysate were detected by SDS polyacrylamide gel electrophoresis (PAGE).Measurements for all of the groups were performed in triplicate, and the values were averaged and reported with their standard deviation (SD). One-way analysis of variance (ANOVA) was performed using the SPSS Program, version 17.0 (SPSS Inc., 2009). The mean values were compared using Dunken Multiple Range Test (DMRT) at p<0.05. Figures were performed through Excel 2010.The antibacterial activities of hen egg white protein hydrolysate (HEWPH) with different time hydrolysis were studied. Radial diffusion assay S. aureus and E. coli showed that hydrolysis during 3-5h had a significantly inhibition zone higher than 0-2h (p<0.05). Flow cytometry analysis of HEWPH at 3 hour hydrolysis for S. Aureus and E. coli were 99.7% and 85.9%, respectively. The hydroxide radical scavenging ability of HEWP reached higher at 3 hour hydrolysis.Quail egg white protein hydrolysate (QEWPH) had a significant high inhibition zone at 3-5h hydrolysis for both bacteria tested (P<0.05). The flow cytometry analysis of QEWPH at 3 hour hydrolysis for S. Aureus and E. coli were 99.8% and 85.7%, respectively. Hydroxide radical scavenging ability of QEWPH at 3 hour hydrolysis was higher than 0-lh.The antibacterial activities of duck egg white protein hydrolysate (DEWPH) with hydrolysis time 3-5h had a significant high inhibition zone. Based on the flow cytometry analysis, DEWPH 3 hour hydrolysis for S. Aureus and E. coli were 99.8% and 85.7%, respectively. Hydroxyl radical scavenging ability of DEWPH 3 hour hydrolysis higher than Oh and 1h.Comparison of three kinds egg white protein hydrolysate showed that the hydrolysate from quail and hen egg is Galliformes order higher than Anseriformes such as duck egg white protein hydrolysate, statistically. Based on types of bacteria, S. Aureus (gram positive) was more sensitive than E. coli (gram negative) for all samples tested. The hydroxyl radical scavenging activity of different egg white protein hydrolysates showed that DEWPH higher than HEWPH and QEWPH.Based SDS PAGE of three kind of sample hydrolysate with high antibacterial activity showed that 10-15 kDa as a major protein number, it is simililar with molecular weight of lysozyme protein.
