The Artemisia sphaerocephala Krasch Polysaccharide(ASP), a traditional Chinese medicine and food additive, is rich in resources. What’s more, it has been reported to have bio-functional property, and it will be more and more important to research the bio-functional property of ASP. In this work, ASP and the high/low molecular weight fraction(60P/60S) were extracted from the outer layer of the Artemisia sphaerocephala Krasch seeds.This experiment will determine the effects of the three kinds of polysaccharide components on peripheral blood mononuclear cells(PBMC) and natural killer cells(NK) in vitro. We will analysis the effects by the determination of activity of proliferation and cytotoxicity. In order to study the differences between the effects of Artemisia sphaerocephala Krasch polysaccharide and Oat beta-Glucanon on the physiological parameters and the structure of gut microbiota, we established Human Flora-Associated(HFA) mice model, which partly simulate the characteristics of the human intestinal flora. The serum biochemical indexes were determined, pathological slices stained with HE of liver and fat were evaluated, and the change of gut microbiota was analyzed by PCR-DGGE(Denaturing Gradient Gel Electrophoresis). The results showed that:(1) Compared with the control group, the proliferation of PBMC in polysaccharide treatment groups showed a trend of increase after decreases first, and the effect of ASP is better than that of 60 P and 60 S. After treatment with 100 mg/L-1000 mg/L of ASP, cell proliferation activity had significant difference(p<0.01). After treatment with 500 mg/L of 60 P, proliferation activity of PBMC significantly enhanced(p<0.05).(2) Compared with the control group, the proliferation of NK cells in polysaccharide treatment groups showed a trend of increase after decreases first, and the effect ASP is better than 60 P and 60S(p>0.05);(3) No significant changes in the killing activity of NK cells were observed between polysaccharide treatment groups and control group.What’s more, there’s no significant difference between polysaccharide treatment groups(p>0.05);(4) Two kinds of polysaccharide in the HFA mice can both reduce serum fasting glucose level(p>0.05) compared with CT group, CT+OG group can significantly reduce the content of total triglyceride(p<0.05). Total cholesterol content and high density lipoprotein cholesterol content in CT+ASP group were significantly higher than that of CT and CT+OG group(p<0.05).(5) CT+OG group significantly decreased the volume of fat cells obviously than CT+ASP group and CT group, and cell arrangement went more closely.(6) Moreover, the structure of gut microbiota in polysaccharide-diet-fed HFA mice changed, their gut bacterial diversity index increased, and the effect of ASP was superior to Oat beta-glucan(p>0.05).
