Edible bird’s nests(EBNs) with abundant nutrition and medicinal value are nests built with saliva and feather by swiftlets in their breeding season. The research on biological activity and test method of EBNs has attracted more and more attention with the consumption increasing in recent years. EBNs are rich in bioactive sialic acids, proteins, carbohydrates, microelements, and water-soluble amino acids. These nutrients can not only be used for anti-aging, anti-viral,anti-radiation, anti-high blood pressure, but also for promoting mitosis and cell regenerating. The current identification methods of EBNs contain sensory testing, physical and chemical, spectral,biological, and chromatographic identifications. This project briefly introduced EBNs in the following aspects: productions, components and detection methods. Then we studied on the identification of EBNs by liquid chromatography-quadrupole-time of flight tandem mass(LC-Q-TOF) and raman spectroscopy.We established a method for identifying EBNs based on LC-Q-TOF combined with chemometrics. The pulverized samples were extracted in 1% formic acid, and TOF mode was used to collect mass spectrums. The mass spectrums were analyzed to filter characteristic compounds and principal component. The cumulative contribution rate of four pricipal components was 94.28%. There were 51 compounds which met the requirement of P<0.05,FC>2 in T test. The prediction accuracy of the method was 100% for training samples and 99% for test samples. Scores of the result was negatively correlated with the percentage of adulteration.This method is suitable for identification of EBNs accurately realizing identification of adulterated degree.We next explored raman spectroscopy method to identify EBNs. The samples were ground and mixed, then raman fingerprint spectrums were collected at the condition of the excitation wavelength of 785 nm. The characteristic peaks of EBNs were 828 cm-1, 1000 cm-1,1237 cm-1, 1330 cm-1, 1448 cm-1, and 1668 cm-1. This peaks did not appear in the adulteration such as starches, agarics and skins. This method is sample, rapid and suitable for identifying EBNs realizing identification of 5% adulteration amount.Five samples were identified with LC-Q-TOF and raman spectroscopy. The correct rate of the two methods were 100%. But, to determine adulterated sample, the method of LC-Q-TOFcan determine the degree of adulteration, and raman technique can determine the type of adulterated. This two kinds of methods could validate and complement each other.
