Magnetic polymer microspheres is a novel functional materials developed in recent years.Magnetic polymer microspheres containing nanoparticles make microspheres have magnetic responsiveness,and the polymer molecular chains can be given specific properties of polymer microspheres by a series of chemically modify.Magnetically targeted drug therapy with high efficiency, less medication, side effects and other characteristics, which is a new method for the treatment of tumors developed in recent years. In this paper, the above problems have been studied, as follows:1. Preparation of PCL microspheresThe preparation of PCL is used by emulsion solvent evaporation method, we check five aspects influence the preparation of polymer microspheres,which were PCL concentration, polyvinyl alcohol (PVA) concentration, the internal oil/water ratio, external oil/water ratio, stirring speed the.By laser granularity instrument, X-ray diffraction (XRD), infrared spectrum (IR), scanning electron microscopy (SEM), the products were characterized, it is concluded that the best conditions of preparation of PCL obtained were PCL concentration was 2%, PVA concentration was 1%, internal oil/water ratio was 1:2, external oil/water ratio was 1:2, stirring speed was 2000 r/min.2. Preparation of CoFe2O4/PCL magnetic microspheresAccording to the best condition of preparation of PCL microspheres, we put magnetic nanoparticles into them in order to prepare magnetic microspheres.By such means as infrared spectrum (IR), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), scanning electron microscope (SEM) checked characterizations of the product.The best preparation of magnetic microspheres have been concluded that magnetic content was 40%.3.Preparation of 5-Fu/CoFe2O4/PCL drug carried magnetic microspheresAccording to the optimum condition for the preparation of magnetic microspheres, we join 5-fluorouracil drug into them to prepare drug carried magnetic microsphere. Through infrared spectrum (IR), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), scanning electron microscope (SEM) and other means of testing, the products have been characterized. A research on how the 5-Fu concentration influence drug-loaded rate and envelopment rate in drug carried magnetic microspheres.With the increase of the amount of 5-Fu, drug loadings showed a trend of decrease after the first increase, the envelopment rate is gradually reduced. The envelopment rate was 27.5%, and drug-loaded rate was29.6%. Finally, we examined the drug release properties, the study found that the magnetic carrier drug microsphere in vitro drug release curve can be divided into three stages:sudden release phase, diffusion phase, the equilibrium phase.When the amount of 5-Fu was 300mg, the product’s percentage of drug release is about 94%.
