Biophotonics is a frontier subject which is formed by advanced photonics and biology techniques. Optical bioimaging is a branch of biophotonics. We make a combination of several nano-materials and advanced optical systems and carry out a series of related work.In this thesis, near-infrared (NIR) fluorescent nanoparticles and upconversion nanoparticles (UCNPs) are introduced. We also describe the methods of their surface functionalization in detail and some optical properties of them were characterized. The nanoparticles can be utilized as efficient probes for bioimaging. We utilize NIR fluorescent nanoparticles as bright optical probes for sentinel lymph node (SLN) mapping of mice, whole-body functional bioimaging and deep-tissue microscopic bioimaging. UCNPs have lots of advantages for bioimaging. We propose a novel strategy to excite UCNPs. Compared with traditional excitation method, we can achieve larger imaging depth. In bioimaging research, the design and building of novel optical systems provide effective tools for imaging. We design and build total internal reflection fluorescence microscopy system, micro-Raman detection system and multi-photon fluorescence microscopic scanning imaging system which provide favourable conditions for further study on bioimaging.
