Plant endophyte is a new type of microbial resources. It is a microorganism that certain stages or all stages of life survive in tissues and organs of healthy plant. The special microorganism and host plant form complex relationship of mutualism and fighting against the enemy. Endophytic fungus has unique metabolism because of their special living environment. Then, it could synthesize many metabolites with novel structures and diversity of activities. Therefore, to find leading compounds from plant endophyte has become one of the most active fields of innovative drug research.Paris polyphylla Smith var. yunnanensis(Franch.) Hand.-Mazz. is a precious herb in Yunnan Province. An endophytic fungus Aspergillus Versicolor was isolated from the rhizome of it, and the chemical constituents of its fermentation products were investigated. As a result, five new novel scaffold of indole alkaloids(1*-5*) and five new isocoumarins(6*-10*), together with eleven known(11-21) compounds were isolated. They are as follows: aspergilline A(1*), aspergilline B(2*), aspergilline C(3*), aspergilline D(4*), aspergilline E(5*), versicolol A(6*), versicolol B(7*), versicoumarin A(8*), versicoumarin B(9*), versicoumarin C(10*), angelicoin A(11), periplanetin D(12),(3S,4S)-dihydroascochin(13), phomolactone B(14), 6,8-dihydroxy-3-hy-dro-xymethylisocoumarin(15), 6,8-dihydroxy-3-methylisochromen(16), 4,6-dihydr-oxy-3,9-dehydromellein(17), fusariumin(18), penicimarin F(19), 8-hydroxy-3-(4-hydroxypentyl)isochroman-1-one(20)、speradine A(21).Compound 1*-5* and 8*-14 were tested for antibobacco mosaic virus(anti-TMV). The results showed that all of compounds have significant activity. In addition, compounds 1*-10* were screened for their cytotoxic activities against the NB4, A549, SHSY5 Y, PC3, and MCF7 human cell lines. It turned out that compounds 1*, 3*, and 4* were found to possess moderate cytotoxicity against all the test cell lines, with IC50 values ranging from 1.2 to 4.2 μM.In this study, we choose simultaneous distillation extraction as a pretreatment to analyse the chemical components of aroma in tobacco leaves through fermentation by Aspergillus Versicolor, an endophytic fungus which was isolated from Paris polyphylla Smith var. yunnanensis(Franch.) Hand.-Mazz.. The results showed that 50 kinds of aroma componenets were detected in the tobacco leaves, and aroma components content of 176.80 μg/g. 64 kinds of aroma components were detected in the tobacco leaves through fermentation, and aroma components content of 378.07 μg/g. Therefore, it could increase the content of chemical components of aroma in tobacco leaves through fermentation by Aspergillus Versicolor.
