Benzodiazepine of polycyclic compounds are an important drugs with a wide range of pharmacological activity and biological activity, and also are a class of compounds which to be first identified privileged structures.The compounds can act on different biological targets, and is a hot spot in chemistry drugs field.Pyridine amines and pyrimidine amines compound and their derivatives are a class of important benzodiazepine with extensive physiological and pharmacological activity and have attracted extensive attention of people.They are commonly used for drugs treating cardiovascular and cerebrovascular diseases, allergies, mental illness and AIDS, a great success has also been made in the treatment of cancer. So the study of the chemical structure and physiological function attracted wide attention from researchers. In this thesis, the raw material are 2- chloro -3- nitro pyridine and 4,6- dichloride -5- nitro pyrimidine and synthetise a series of pyridine and pyrimidine amines compounds.The first chapter summarizes the physiological and pharmacological activity of benzodiazepine,introduces several kinds of organic oxidation reactions. Then described the air or oxygen classification and its current oxidation situation, we also introduced the oxidation of aromatic side chain ang methylene,described the definition of Bischler-Napeiralski cyclization reaction and its application in organic synthesis.,and respectively introduced physiological activity and application of pyridylamino and pyrimidinamine, and gives the significance of the research.The second chapter is mainly the experimental section. Taking 2-chloro-3-nitro pyridine and 4,6-dichloro-5-nitro pyrimidine as reactants to synthesis the precursors of Bischler-Napiralski ring closing reaction, namely N2-methyl-N2-phenyl pyridine (2,3) diamine and 5-amino-4-(N-methyl phenylamino)-6-chloro pyrimidine. Then they respectively respect with propanoic acid, butyric acid, phenylacetic acid to finish the ring closing and oxidation reaction. We choose the optimal condition for the oxidation reaction: pyridylamino using acetonitrile as the solvent, the diatomite as carrier has the best oxidation effect; pyrimethamine in the solvent of THF, activated carbon as the carrier, has the ideal oxidation. Products were analysized by 1H-NMR and mass spectrogram.In chapter three, it described a investigated experiment. Taking o- fluoronitrobenzene or o-nitrochloronemzene as the raw material to synthesize N1- methyl -N1-phenylbenzene(1,2)diainine. Several alkali, such as NaH, KOH, ethanol sodium and two kind of amine:aniline and N-methyl aniline can not reach the goal.
