In this paper, two kinds of hydrophilic interaction chromatography (HILIC)and ion exchange chromatography (IEC) chromatographic separation materials were synthesized based on monodisperse polyglycidylmethacrylate beads (PGMA/EDMA) as matrix by using atom transfer radical polymerization (ATRP) method,methacryloyloxyethyl trimethyl ammonium chloride were grafted on the surface of the substrate; two kinds of hydrophilic interaction chromatography (HILIC) and ion exchange chromatography (IEC) Chromatographic separation materials were synthesized by the chemical bond based on monodisperse polyglycidylmethacrylate beads (PGMA/EDMA) as matrix, anedioic anhydride were grafted on the surface of the substrate. The chromatographic properties of those stationary phases were investigation in details. The separation materials were used to separate different types of protein and nucleoside, and applied it to detected the active ingredients nucleosides in drug Ning xinbao,principal constituent lysozyme in chicken egg white. This paper mainly consists of the following sections:1. A new and original separation material for strong anion exchange (SAX) and hydrophilic interaction chromatographic (HILIC) stationary phase was prepared by atom transfer radical polymerization (ATRP). Methacryloxyethyltrimethyl ammonium chloride was grafted onto the surface of the beads with self-restraint (8.0μm) monodisperse polyglycidylmethacrylate beads (PGMA/EDMA).The poly (Methacryloxyethyltrimethyl ammonium chloride)-grafted beads (ATRP1、ATRP2 and ATRP3) obtained with different ATRP formulations were tried as chromatographic packings in ion-exchange chromatography by changing monomer/initiator ratios/Polymerization degree. Inspected this padding in detail to the standard protein separation performance and the nuclear glucoside separation situation.(1) Inspected this padding in detail to the standard protein separation performance, investigated various factors influencing the behavior of proteins of reservations, includeing:pH, flow rate of mobile phase, and the effect of flow rate on column. Also inspected the polyreaction process, the monomer stem grafting rate and the polymer to the characteristic protein adsorption performance The results showed that three different degree of polymerization of chromatographic packing good separation performance, proteins are the baseline separation of four kinds of standards, and the retention time of protein had a positive performance with increasing the chain lengths of the grafted poly (Methacryloxyethyltrimethyl ammonium chloride).(2) Inspected ATRP3 packings in detail to the standard nuclear glucoside HILIC chromatographic fractionation performance, also investigated various factors influencing the behavior of nucleotides of reservations, includeing:pH, salt concentration, water content. The results showed that ATRP3 packings could also be used in HILIC, seven kinds of nucleoside compounds can baseline separation. Finally, the separator material was used for determination of the active ingredients nucleoside compounds in drug Ning xinbao. Good results have been achieved.2. Two kinds of hydrophilic interaction chromatographic (HILIC) and weak cation exchange chromatography (WCX)chromatographic stationary phases were synthesized based on self-restraint (8.0μm) monodisperse polyglycidylmethacrylate beads (PGMA/EDMA) as matrix. The stationary phase was evaluated in details to determine its separation performance of standard protein and nucleoside.(1) Inspected this padding in detail to the standard protein separation performance, investigated various factors influencing the behavior of proteins of reservations, includeing:pH, Flow rate of mobile phase, and the effect of flow rate on column. Finally, the separation material was used to determination of the principal constituent lysozyme from egg white. The results showed that the prepared columns could separate six kind of tested nucleoside compounds, polymerization of chromatographic packing good separation performance.
