Xylanase is a composite class of enzymes belong to hydrolases. Xylan is a kind of hemicellulose, rich in plant, which is predominantly linked by β-1,4-xylose units in cellulose. Xylan is one of the most significant recyclable recourses. Xylanase can hydrolysis of xylan from plant raw materials, feed into monosaccharides or oligosaccharides. At present, the xylanase is recognized in the world, which is one of the most promising green, environmental protection enzymes. In this study, xylanase used in stover for ethanol production was as target, and explored a number of key technologies in the xylanase fermentation production processes, moreover developed a new solid-state fermentation equipment. It will solve the key problems in the production of xylanase production, such as screening technology, fermentation technology, research of solid-state fermentation equipment. The results are as follows:The high-yield strain:Aspergillus (M-Asp-S1), which is suitable for degradation straw fiber raw materials, was screened out, and the culture conditions were optimized. The solid-state fermentation xylanase production medium was obtained by single factor and orthogonal test:70% corn cobs and 30% bran,4% (NH4)2SO4, KH2PO4 0.75%, Mg2SO4 . 7H2O 1.0%, Propylene glycol 0.10%, water 70%. Enzyme activity increased 3.44 times from the original 900.61 U/g to 4000.34 U/g. The new solid-state fermentation equipment was successfully established and the production processes was optimized. The liquid seed vaccination and solid-state fermentation were used in the production process. With this new type of continuous culture equipment, enzyme hydrolysis was quickly, and the yield of monosaccharide was high, and the utilization rate of raw materials increased by 20%. By comparing the enzymes composition and enzyme activity of domestic companies, the composition of xylanase studied in this essay was more reasonable. The effect of enzymatic hydrolysis and fermentation were good. To sum up, the breakthrough of this study’s key technical for the large-scale promotion of straw fiber ethanol production and application model was established.
