China is the biggest egg production and consumption country in the world. Recentyears, egg processing industry has developed a lot and become an important part infood processing. Eggshell is the most important by-product in egg processing. Withthe development of egg processing industry, the comprehensive utilization of eggshellhas become the inextricable problem. The environment pollution and the resourcewaste caused by eggshell has been the bottleneck problem in egg processing. Eggshellis a good calcium resource. Calcium has a great meaning on maintaining bone healthand other human body normal physiological activities. Compared with inorganiccalcium salt and organic acid calcium preparations, calcium chelating amino acid hasbetter chemical and biochemical stability. Calcium chelating amino acid is easy to bedigested and has double effects including calcium supplement and amino acidsupplement. Calcium chelating amino acidis the future development direction ofcalcium production. This project is based on the low efficiency of eggshell problem,combined with itself characteristics and with the aim of developing safe functionalcalcium chelating amino acid.Chelate ratio and in vivo efficacy of calcium chelatingamino acid were observed. Calcium chelating phenylalanine was selected as thetypical example of developing calcium chelating amino acid from eggshell and thefurther research on toxicology and physical nature was carried out. This projectprovides theoretical foundation and technical guidance for developing calciumsupplement production from eggshell.This paper is supported by Project of National Key Technology Research andDevelopment Program for The12th Five-year Plan(NO.2012BAD28B08). Thisarticle main research content was as follows:In two kinds of calcium chelating amino acid research, chelate ratio was regard asthe index. Comprehensive compared chelating time, reaction temperature, liquid-solid ratio and material ratio effect of calcium chelating phenylalanine and calciumchelating methionine, the optimized condition was got by the ternary quadraticorthogonal optimization.The optimized condition of calcium chelating phenylalanine was as follows:chelating time was73min, reaction temperature was35℃and material ratio was2.7:1(g/g). The optimized condition of calcium chelating methionine was as follows:chelating time was73min, reaction temperature was36℃and material ratio was2:1(g/g). The chelate ratio of calcium chelating phenylalanine was93.51±0.02%. Thechelate ratio of calcium chelating methionine was92.53±0.01%. This result showedthat the chelation of calcium chelating phenylalanine was better than calciumchelating methionine.In the research of comparing the calcium abortion effect in vivo of different calciumsupplement, distilled water, calcium gluconate, calcium chelating phenylalanine andcalcium chelating methionine were choose. The serum calcium content, serumphosphorus content and serum alkaline phosphatase (ALP) content of Kunming malemice were identified to determine the calcium abortion effect of calcium chelatingamino acid. The results showed that both calcium chelating phenylalanine andcalcium chelating methionine had well ability of adjusting the serum calcium content,serum phosphorus content and serum alkaline phosphatase (ALP) content and hadeffect on organic matter content of the bone. The results showed that calciumchelating phenylalanine had better calcium abortion effect than calcium chelatingmethionine. The optimal dose of calcium chelating phenylalanine was1200mg/(kg d).In the research of calcium chelating phenylalanine powder’s toxicologicalevaluation, mice weight, food consumption, water consumption and organ coefficientwere investigated. The results showed that, compared with distilled water, differentdose of calcium chelating phenylalanine had significant improvement of mice spermaberration rate and bone marrow micronucleus rate (P<0.05). Compared with glucosesour calcium, calcium chelating phenylalanine had no significant improvement ofthese indexes (P>0.05). Different dose of calcium chelating phenylalanine had no significant improvement of these indexes each other (P>0.05).In the research of calcium chelating phenylalanine powder had effect on miceweight, food consumption, water consumption and organ coefficient, the resultsshowed that calcium chelating phenylalanine had no effect on mice. This meanscalcium chelating phenylalanine had no side effects.In the research of physical nature of calcium chelating phenylalanine, thewater-retaining property, hygroscopicity and oil absorbency was observed. The resultsshowed that the water-retaining property was68.3%and oil absorbency was5.3mL/g.The hygroscopicity of calcium chelating phenylalanine under R.H43%was37.2%,under R.H81%was65.3%. Microbiological testing by E. coli, Salmonella, the totalnumber of colonies found phenylalanine calcium chelate complex national standards.It is a safe food.This project indicated that calcium chelating phenylalanine is a high quality andsafety calcium supplement and provides theoretical foundation and technical guidancefor developing calcium chelating methionine.
