Dioscorea alata Lirm is a year or perennial liana, and it’s belonged to the Dioscoreaceae,Dioscoreacea L. The distribution of Dioscorea alata Lirm has obvious regionality, mainly produced in south of Yangtze River and Yichun, Jiangxi province is one of major production. Resistant starch can regulate blood sugar, weight control, improve intestinal health and so on. Dioscorea alata Lirm starch was prepared RS3 as raw material. In this study, RS3 was prepared by the combination of ultrasonic and complex enzymes, in order to improve the yield of RS. Furthermore, RS was added to soda cracker to study its effects on the quality of soda cracker, to determine the best formula of soda cracker containing resistant starch. The main research contents and results were as follows:1. Single factor experiments and response surface experiment were performed to optimize the conditions of ultrasonic treatment. Each factor affect ing the yield of RS are ranked as follows: the concentration of starch﹥ultrasonic outpower﹥treatment time. The results showed that the optimal conditions were that the co ncentration of starch 7.0%(W/W), ultrasonic outpower 225 W and treatment time 50 min. The yield of RS was 12.42%.2. In order to optimize the preparation conditions of resistant starch, single factor and response surface experiments were performed to opt imize the conditions of enzyme solutions and it was performed based on the optimal ultrasonic conditions as described above before. Each factor affecting the yield of RS are ranked as follows: pullulanase amount ﹥ effect time of thermostable α-amylase ﹥ effect time of pullulanase﹥thermostable α-amylase amount. The results showed that the optimal conditions were as follows: thermostable α-amylase 4 U/g dry starch, the enzymatic hydrolysis time 16 min, pullulanase 9 U/g dry starch, the time of treatment by pullulanase 3.6 h. The yield of RS was 18.57%.3. RS was added to soda cracker to establish the best formula of soda cracker with resistant starch by single factor and response surface experiments. The results showed that the best formula was: flour 76 g, RS 24 g, oil 31 m L, emulsifier 10 g, milk 60 m L, salt 1 g, 200°C baked 8 min. In a mouse model, the soda cracker was as effective as diformin tablet in terms of easing the weight loss and postpra ndial hyper-glycemia.
