Polysaccharides in animals and plants and microorganisms have various bioactivities, such as anti-tumor, reducing blood sugar and blood fat, adjusting immune function and antioxidationt. Auricularia Auricular polysaccharide, a physiological active substance, was very hot recently. Chromium is an essential trace element in human body. Meanwhile, it is the necessary element for the hypoglycemic effects developed by insulin. Chromium (III), as the effective element of glucose tolerance factor, has the function of ameliorating glucose metabolism for diabetics. This topic will both together effectively, making further research on subsidiary treatment for subsidiary treatment for hyperglycemia.The main content of this study is described as follows. Specific research results are as follows:1.The inhibitory effects of these extracts to α-glucosidase activity were investigated by in vitro enzymatic method. Research on the different extracts of Auricularia auricular polysaccharides to α-gucosidase inhibition in vitro and different segments of alcohol sink inhibition rate on α-gucosidase.Auricularia auricular polysaccharides extractive and different alcohol precipitation fragments have an inhibiting effect on α-gucosidase activity, and they showed obviously concentration-effect curve relationship. Auricularia auricular polysaccharides have the inhibiting effect on α-gucosidase. Three different solubility of Auricularia auricular polysaccharides samples. Among them, the inhibitary activity of acid Auricularia auricular polysaccharides is stronger than others.On the different acid Auricularia auricular polysaccharides alcohol precipitation fragment, the inhibitary activity of 80%alcohol precipitation fragment is stronger than others. In the different segments by NaCl eluting, the inhibitary activity of 0.2mol/LNaCl eluting fragment is stronger than others. Thus it is concluded that it is the elected sample and named it AAP.2.Previous studies have found that sulfate esterification Auricularia auricular acidic polysaccharides has good biological activity.Response surface methodology was adopted to optimize sulfuric acid esterification process conditions for Auricularia auricular polysaccharides. Studied the volume ratio of chlorosulfonic acid and N, N dimethyl formamide, the volume ratio of esterification reagent, the reaction temperature, the reaction time, rotational speed five factors influence on degree of substitution of the ulfatex acid esterification for polysaccharide of Auricularia auricular. The path of steepest ascent was used to define optimal response region for these three factors. Optimal level combinations of factors were obtained through Box-Behnken design. The optimal sulfuric acid esterification process conditions for polysaccharide of Auricularia auricular were as follows:the volume ratio of chlorosulfonic acid and N. N dimethyl formamide is 1:3.49、the volume ratio of the esterification reagent is 33%% the reaction temperature is 50℃、the reaction time is 112.84min、rotational speed is 200r/min.Under optimal sulfuric acid esterification conditions, the DS of polysaccharide reached 0.4905. The FTIR analysis established that the SAP had specific absorbing peaks of -OSO3 and C-O-SO3 at both 1249 cm-1 (S=O) and 799 cm-1 (C-O-S).3.The effects of three independent variables (The quality of sulfate esterification Auricularia auricular acidic polysaccharides and chromium(Ⅲ),chelating time, chelating temperature, pH and chromium(III) concentration of initial) with spectrophotometric method. The optimal sulfuric acid esterification process conditions for polysaccharide of Auricularia auricular and chromiurn(III) were as follows:the quality of sulfate esterification Auricularia auricular acidic polysaccharides and chromium(Ⅲ) is 3.68:1, Chelating temperature is 55.72℃, Chelating time is 3h. Chromium(III) concentration of initial is 5.77 mg/mL, the pH value is 5. Under the optimized conditions, chelating rate is 24.00%. It is feasible to optimize the chelation processing of SAP-Cr. The FTIR analysis established that the SAP had Forming a chelate.Given that, it could provide several references for further research on detection of it.4.The AAP and SAP-Cr was used to treat rat models of diabetes induced by alloxan.The changes in body weight, fasting blood glucose (FBG), hexokinase(HK), succinic dehydrogenase (SDH) were determined in diabetic rats before and after the treatment. Animal experimental results show that:by in vivo hypoglycemic results, The AAP and SAP-Cr both could significantly slow down the negative growth of body weights in diabetic rats, and significantly increase HK, SDH.The AAP and SAP-Cr was positive in subsidiary treatment for the fasting glucose.Results showed that AAP and SAP-Cr had significantly hypoglycemic effect compared with the model group.
