In recent years, with the rapid development of Industry and agriculture, more chemical fertilizers were added to the soil. This phenomenon has led to declining soil fertility and soil structure damage, the rapid development of industry causing serious heavy metal pollution. So versatile high potency probiotic research has a very important economic and social significance.This study did research in the following areas:First, we activated Bacillus mucilaginosus which was kept in our laboratory,then we identified it morphological and molecular biology and studied some of its properties, we named it Bacillus mucilaginosus G14; we did market researchs and collected nine products of Bacillus mucilaginosus. We did Shake flask fermentation experiments, flowerpot soil colonization experiments, potassium test solution, and analyzed the development potential of Bacillus mucilaginosus G14.Secondly, we have chosen Bacillus mucilaginosus G14 as the starting strain, optimized the conditions of the medium and fermentation conditions on the level of flask. Optimizing experiments showed:When 1.0%molasses dosage,0.5% nitrogen dosage, initial pH 7.0, fermentation temperature is 30℃, the strongest capacity of fermentation was 3.64 X 108 cfu/mL. On this basis, we did the 50 L pilot fermentation tests, fermentation inoculum, logarithmic phase of dissolved oxygen, spores mature and yield optimized, The results showed that:12% inoculation volume met the the fermentation requirements, a higher inoculum is useless; we optimized ventilation logarithmic phase, when the guarantee of DO was 50 ± 5%, the logarithm of the smooth, stable and enrichment enrichment achieve the best results, the lowest rate since the dissolution, OD value reached 18.5; we optimization of Bacillus maturation and ripening time, pH has a significant effect on the formation of spores, in the stationary phase when the pH was adjusted to 8.5, spores can reach more than 95% yield and to shorten the time of sporulation to 8.5 hours; Finally, the batch fermentation tank, worked out the optimal fermentation parameters. After the fermentation products were analyzed preservation, explores the impact of the added amount of sodium benzoate and pH on the preservation of time, and at room temperature preservation sinensis preparation time, The results showed:sodium benzoate dosage 0.3‰, pH was adjusted to 2.5, the liquid formulation can reach 20 days of storage times. The powder formulations at room temperature preservation, the first five months after the reduction to 50%. Basic demise 8 months later.Then we explored the Bacillus mucilaginosus G14 used in the heavy metal chromium sewage treatment, optimizing the production of adsorbent media and adsorption kinetics analysis, The results showed:Under nitrogen 0.3% addition level of culture conditions, the ability of microorganisms producing glial G14 strongest adsorbent. Biosorption of Cr (Ⅵ) from aqueous solution by Bacillus mucilaginosus G14 was investigated, and the effects of pH, initial concentrations of Cr (Ⅵ), adsorption time, and temperature were studied. The experimental results showed that:the optimum biosorption can be reached in 20 minute under the conditionsof T=20℃, pH= 5,Cr=20 mg/L, more than 80% of Cr(Ⅵ) was adsorbed and the maximum adsorption capacity was 257 mg/L. The experimental data was well fitted by the Langmuir model. Analysis of FTIR(Fourier Transform Infrared Spectroscopy) showed that functional groups and surface structure changed after adsorption.Finally, we studied the gum used in the chili powder’ greenhouses, by experiment shows that when the dosage 10 × 109 cfu/m2, the struggle to achieve the best production results, an increase of about 1000 m2 per 365 kg, can be about 1000 yuan of net profit, add the amount is too low and too high are not better value for money.
