Succinic acid is an important product of the citric acid cycle. It is widely present in the human body, animals and plants, and microorganisms. It is also important intermediates of many compounds. At present, the method for the synthesis of succinic acid is mainly chemical synthesis, biotransformation and microbial fermentation. By microbial fermentation method has the advantages of low cost of raw materials, mild reaction conditions, therefore, the use of microbial fermentation recently becomes an important method for the synthesis of succinic acid. However, the efficiency of extraction, concentrated, acidified and the purification process obtained by fermentation of succinic acid is very lowly and high costs. So to achieve that fermentation-based production process has a certain degree of difficulty. In this paper, study several key aspects of succinic acid fermentation extract and examine various aspects of the optimum parameters of the key factors, some details are as follows:1 By comparison, select Ca Cl2 as calcification agent. Through four factors of affecting the process of calcification for example concentrations of calcification agent Ca Cl2, different the reactants, reaction time and reaction temperature four single factor experiment and orthogonal experiment. Obtain the best conditions for calcification is 40% of concentrations of calcification agent Ca Cl2, the reactants as 1:1.4, reaction time is 30 min, reaction temperature is 75℃.2 Through the study of acid hydrolysis process two factors for example sulfuric acid concentration and temperature, obtain sulfuric acid concentration of 60%, the reaction temperature is 80 ℃for the best conditions succinic acid hydrolysis of calcium.3 Through the absorbance at different wavelengths fermented corn syrup extract study results at 410 nm for fermentation extract optimal wavelength. Through the study of several decolorizing material’s decolorization rate and loss rate of succinate, select TX-328 P as corn syrup fermentation extract bleaching material, the decolorizing material decolorization corn fermentation extract up to 90%, and the loss of succinic acid rate was very low. In addition, studies four factors of decolorization process as p H, bleaching time, bleaching temperature and amount of activated carbon, no need to adjust the p H, and decolorization time was 30 min is the best decolorization, bleaching temperature is 80℃, the amount of activated carbon is 0.8%.4 By study reationship between several different anionic resin and capacity of ion-exchange Ion exchange capacity of 330, D315 and D301 are seldom, they are the type of resin required for the experiment. By comparison, D315 is the best, the maximum yield of succinic acid using D315 resin 97.8%. Then study the effect of temperature on the column and the column flow yield of succinic acid, the optimal temperature of ion-exchange desalting column is 25℃, the best flow rate of column flow is 2BV/h. |
