Water keeps all the animals on the earth, and it is the necessary condition of the development of human. With the rapid development of economy, the increasing population, water resources suffered more and more serious pollution.It makes the water resources supply and demand more urgent. Phenol containing wastewater is most common, it is of great harm and pollution range. Phenol is a possible carcinogen recognized residue, high concentrations of phenols in water to human, animal and plant has a direct harm. Therefore, green, efficient phenol containing wastewater treatment technology has become a difficult problem that governments of various countries and scientists need to solve. Compared with other processing technology, biological enzyme as an efficient catalyst, can directly effect of chemical chain of organic pollution.It has some advantages, such as mild condition, high efficiency, low cost and no toxic side products,etc.It has gradually become a focus of water pollution processing of application and research.In this paper, immobilization on the porous polystyrene nano-fibrous membrane of horseradish peroxidase(HRP) were prepared by electrospinning method, and for the phenolic wastewater treatment focuses on the electrospinning parameters on the fiber morphology, effect of immobilized enzyme conditions on HRP enzyme activity,and the properties of immobilized HRP for treatment of wastewater containing phenol. The research of this project has important significance to the development of new enzyme immobilization carrier materials and innovative wastewater treatment technology.Correlative researches are listed as follow:(1) Research on the electrospinning and it’s the best spinning process for preparation of polystyrene nano-fiber membrane.In order to obtain the best effect of electrospun jet, and the best polystyrene nano- fibrous morphology the influence factor were discussed through optimizing the electrospinning conditions, such as solvent polarity and ratio, environment humidity and concentration of polymer and so on. The optimum parameters of electrospun fiber membrane as follows: polystyrene concentration was 15%(wt), solvent(DMF:THF) ratio was 50:50, voltage was 16 k V, the receiving distance was 15 cm.(2) Studied on the electrospun polystyrene nano-fiber HRP immobilized enzyme and optimal immobilization parameters.Coupling with surface adsorption method, immobilized enzyme on the surface of polystyrene nano-fiber after the change traits. Studied on the effect of immobilization time, enzyme concentration and PH parameters of HRP immobilized enzyme activity. The resultsshowed that the optimal HRP immobilized enzyme conditions are as follows: fixed time was 4 h, PH 7.0, enzyme concentration was 200 U/L. Further studies show that, HRP enzyme immobilized compared to the free enzyme, placing stability, thermal stability and PH stability were greatly improved.(3) Studies on the performance and optimization of HRP enzyme immobilized and treatment performance of phenol.The treatment effect on phenol adsorption, the pure physical contrastive study of polystyrene nano-fiber membrane free enzyme and polystyrene nano-fiber membrane immobilized enzyme under three conditions. The results showed that the catalytic effect on the treatment of phenol was significantly higher than that of the immobilized enzyme membrane physical adsorption and free enzyme. The optimum conditions for further investigation on the treatment of phenol by immobilized enzyme, phenol removal rate is the highest when the immobilized enzyme dosage was 0.25 mg/g, the phenol concentration of 75 mg/L, molar ratio of hydrogen peroxide and phenol was 1:1.
