Non-Saccharomyces yeasts laid an important foundation for wine aroma components and promoted the development of the wine industry. However, domestic wineries making wine with yeast essentially imported, this not only is a waste of yeast resources, but also has certain effects on the biodiversity of local yeast. The research and utilization of Non-Saccharomyces yeast is still in the initial stage in our country,and has a big gap compared with foreign. Focus of the study is screening non-Saccharomyces cerevisiae with more competitive in Xinjiang favorable geographical conditions and raw material resources.In this study, 250 yeast strains has been separated from the must in vineyard, in Shihezi 141 tuan,143tuan,145 tuan,147tuan,152 tuan. Morphological identification,initially divided into five categories, fermentation tolerance test of representative strains, has strong tolerance in high concentration of alcohol, high SO2 concentration and high-sugar, some strains has good fermentation performance in alcohol was 14%,SO2 concentration of 400mg/L of liquid fermentation medium in fermentation. Well tolerated by study of two strains of a variety of physical and chemical indicators,domesticated, get high-quality wine yeast resources, Y16 and Y19 genus Pichia.By morphological identification and classification of 21 representative strains,DNA extraction and PCR amplification technology, 26 S r DNAD1/D2 region sequence, following conclusions:Strains belonging to 6 genera: Candida, also Montessori season Saccharomyces(Meyerozyma), red yeast genera(Rhodotorula), Alternaria genus Hansenula(Hanseniaspora), yeast(Saccharomyces cerevisiae) and Pichia, a total of 10 kinds,including six suspected species.The study found that the yeast strains with Gen Bank sequence homology existing pattern of low 6,Y2 and Galacto-mycesgeotrichum JN083822.1 homology was 98%,Y3 Rhodotorulamucilaginosa GU373744.1 homology was 97%, Y4 and Meyerozymaguilliermondii HM98 8720.1 homology was 98%, Y5 and Candida amapae EU057556.1 homology was 98%, Y6 and Y7 and Han seniasporauvarum GU080043.1, HM988683.1 homology was 98% and 97%, etc.According to Kurtzman yeast species division basis, a similar rate of less than 99% homology, and can not be classified as the same species, so for these strains can be identified initially determined that it is a new species, pending further morphology, mode of reproduction and identification of physiological and biochemical research results.
