| OBJECTIVE To establish a method of the determination of the active constituent puerarin in Health Pill and Gegen crude drug and magnolol hi Health Pill and Cortex Magnoliae Officinalis.METHODS A simplified HPLC procedure was developed .The effects of sample preparation,the mobile phase and detection wave length on the separation of the various components hi samples were investigated.A 4.6mm* 150mm Hypersil ODS column with a mobile phase consisting of methanol-H2O(30:70) was used .It was detected at 252nm and the other mobile phase consisting of methanol-H2O(77:23) was used.lt was detected at 294nm.RESULTS Puerarin and other components were well separated.The standard curve was linear over the concentration range of 4 X 10-3 ~ 2.1 X 10~2mg/ml and the correlation coeffient was 0.99826;Magnolol and other components were well seprated as well.The standard curve was linear over the concentrationrange of 8 X 10~2~8 X 10"1 and the correlation coefficent was 0.9979.The recoveries of puerarin were 99.34% for Health Pill and 99.54% for Gegen crude drug and the recoveries of magnolol were 99.46% for Health Pill and 97.33% for cortex Magnoliae officinalis with RSD less than 1.78%.Health Pill could be directly chromatographed after it was diluted by CH3OH and filtrated. CONCLUSION The proposed method was simple , accurate and precise. |
