Chthamalus challengeriHoek is an important species , which was usually used for fouling and anti-fouling study. Since larva is more sensitive to the pollution and environmental stress, Chthamaluschallengeri larva ( naupli II ,naupli VI and cyprid ) was took as experiment material, to study its responses to reactive oxygen species(ROS). Meanwhile ,Fe3 +-EDTA / Vc(ascorbic acid) system was used to generate hydroxyl radical ( OH), and X(xanthine)/XO(xanthine oxidase) system is used to generate superoxide radical ( O2) .The results could not only reveal the damaged mechanism of poisonous pollutants and other environmental stress to organisms , but also offer information for developing non-poison anti-fouling-coating and protecting marine environment. Experiment results showed as follows:1 Two kinds of reactive oxygen species exhibited remarkable damage to larvae on 3 types of different development stage.The treatment of reactive oxygen species could result in decrease of larva survival rate and inhibit metamorphosis rate of cyprid. The larvae on different development stage indicated different sensitivity to reactive oxygen species, the napuli VI showed the most sensitive. 2 Antioxidants could decrease the damage effect of reactive oxygen species on larvae in certain level.The addition of antioxidants in culture system had risen larva suvival rate and the metamorphosis rate of cyprid .The reaction of different antioxidants was different to larvae on different development stage. 3, Peroxidase(POD) and superoxide dismutase(SOD) activity of larvae on different development stage showed difference responses to ROS stress. Compared with control group, POD and SOD activity of the naupli n and naupli VI drops obviously under ROS.stress, while the changes of POD and SOD activity in cyprid were not notable.4 Under the stress of reactive oxygen species ,The changes of MDA content in 3 types of larvae on different development stage were coincidentally opposite tothe changes of their SOD activity. Compared with control group, the MDA content of the naupii VI substantially increased( P < 0.01); the MDA content of the naupii n also showed increment (P < 0.05 ); And cyprid MDA content was not sensitive to reactive oxygen species, it did not change fairly comparing with control group(P>0.05).Therefore,the membrane lipid peroxidation of larvae on differene development stage was different.
