Extraction flavonoids from Hemerocillis citrina Baroni and the extracting technological conditions as well as the separation and purification of flavonoids extracted were studied in this paper. Flavonoids was proved to be flavonol by chemical methods and spectrum(UV ,IR and HPLC). The polarographical characters of flavonoids were investigated by electrochemical methods. A new system for scavenging radicals by electrochemical methods was established at the same time. Dififerent flavonoids show different scavenging action in the process of radical scavenged.1. Hemerocillis citrina Baroni is a Chinese traditional foliage plant which has nourishing components including vegetable alkalis, glycosides, flavonoids, carrot, vitamin, volatile oil, tannins and abio-mineral. During the process of extracting flavonoids from hemerocillis, traditional extracting methods and supersonic extraction were compared and the conditions of supersonic extracting were optimized by vertical-method. The best conditions for the extracting flavonoids from Hemerocillis citrina Baroni were as follows: the Hemerocillis citrina Baroni go through sixty type boult and power of supersonic was midst and extraction time was 20 min and the solid-liquid leaching was 1:8. At the same time, all kinds of factors for effects of extracting in chosen conditions can concluded that the sequence of facters is: powder of sample >supersonic power > extraction time>dosage of solution.2. After extracting coarse solution of flavonoids, adsorption and separation by AB-8 resin, NKA-9 resin, X-5 resin were studied, the best conditions were: resin with AB-8 resin and 70% ethanol as eluted soluent. Flavonoids were proved to be flavonol by chemical methods and spectrum.3. The polarographical characters of flavonoids were investigated by electrochemical methods. The product showed a peak potential of-1.34V (vs, SCE ). That is to say flavonoids have better adsorption by cyclic voltametry and the pole reaction was irreversible. The system of anti-superoxide action radical and hydroxyl radical by electrochemicalmethod were established. It was come to conclusion that flavonoids is an excellent scavengers after comparing eliminating effects of different kind of flavonoids to anti-superoxide action radical and hydroxyl radical, the conclusion can be obtained that the structure of flavonoids have connection with eliminating effects of anti-superoxide action radical and hydroxyl radical. The scavenger of the free radical by electrochemical methods which required inexpensive instrument and high sensitivity and low concentration sample.
