Volatile Oil Extraction And Separation Of Its Antibacterial And Anti-tumor Effect

Zanthoxylum Bungeanum Maxim is a shrub species of Zanthoxylum Linn , Rutaceae, which is a common species in our country. The fruits is one kind of traditional condiments, as well as one kind of famous traditional Chinese medicines. It is used cure vomit, diarrhea, ascariasis etc. And also be used externaiy to eczema and prurigo. Volatile oil is one of the mostly effective active components of the species, here we adopted two methods, supercritical CO2 fluid extraction(CO2-SFE) and ethanol reflux extraction, to extract volatile oil, used the material of the cv. sp. of "Dahongpao" from Shanxi province. Then purified to 9 samples and given further studied on their pharmacological effect of antibioltic and anti-tumor activities.The results of CO2-SFE show that the primary relation of three influential factors pressure (P), temperature (7) and extraction time(t) is: P>T>t. The optimal condition is P 33 Mpa, T 55 C and t 1. 0 h when the material' s 40 order size, 200g. The extraction rates of volatile oil is 10.2%. About 31 kinds of constituents are identified in the volatile oil by GC-MS analysis. The major chemical constituents are 3-Methyl-2-azafluorene, accounts for 18.63%, y -terpinene and 3-carene.We adopted filter paper mensuration and tube culture to estimate its antibioltic activity among 9 samples to 8 kinds of tested germs in the antibioltic experiment. The results show that to bacterial 5.9 has the most obvious inhibition to Escherichia coli, MIC is 1. 25%; S. 8 has the most obvious inhibition to Bacillus subtilis, MIC is 0. 625%;no sample has the most obvious inhibition to Staphylococcus aureus. To fungi, S. 6 has the most obvious spore generation' s inhibition to Aspergillus oryzae3042 and Aspergillus niger, MIC are both 1. 25% S. 1 and S. 9 have the most obvious inhibi tion to Phyllosticta maydis, MIC are both 1.25%; S. 1 has the most obvious inhibition to Colletotrichum musae,MIC is 1.25%; S.6 has inhibition to Saccgaromyces cerevisiae, MIC is 5%. We found that S. 6 has spore generation' s inhibitionto fungi first time, but its effect principle deserves to further research. The volatile oil has obvious antibioltic effect: volatile oil from Z (CO2-SFE) inhibit mainly to fungi, while volatile oil from Z. (alcohol reflux extraction) inhibit chiefly to bacteria.In the rudimentary research in antitumoral effect of the volatile oil, we selected three indexes , observation of cellular form, quantity of vital cell(MTT method measures the enzyme activity of cell)and velocity cellular growth. The results indicate:5. 9 has strong inhibition and kill and wound effect on melanoma cell A375 when its concentration is 1. 5mg/ml, in the same concentration it has no obvious inhibition effect on normal facilitate-fibers cell CCD-39. Because there are only three indexes in our experiment, the research in this field still need further verification.
