The thesis includes the following two sections:Section I: IntroductionThe instrument, principle of high-speed countercurrent chromatograhy were described and the applications for plant, traditional Chinese medicine, protein, food, inorangic compound, chiral compound, et al were introduced. Now high-speed countercurrent Chromatography has become an important preparative separation method.Section II: Experiment1: The separations of coumarin of Radix Angelicae Biserrataae, Radix Angelieae Dahurcae, Radix Peucedani Dahurcae and Herba Artemiside Annuae were carried out with the solvent system composed of CHCl3/CH3OH/H2O=4:x:2(x=2.5~4.5) , which further verified it was a versatile separation system for active ingredient of plant.2. Six hydrophilic and two lipophilic β-cyclodextrins derivatives were synthesized and these derivatives have been used for the chiral additive reagent of high-speed countercurrent Chromatography. We tested fifteen racemic pharmaceuticals and found that carboxymethly-6-cyclodextrins can separate Aminoglutethimide and Chlorpheniramine. Therefore, high-speed countercurrent Chromatography (HSCCC) may play an important role in the chiral separation.
