Develop clinical kits for measuring high-density & low-density lipoprotein cholesterol in serum without prior separation. Detergents and PEG modified enzyme were the key to break lipoprotein selectively. Surfactants whose HLB is between 10 and 13 were found to break lipoprotein more efficiently. From which, NUST-6 was found to break high-density lipoprotein and NUST-A was found to break low-density lipoprotein; When cholesterol esterase was modified with mono-chain or double-chain PEG derivatives, it showed selective catalytic activities towards HDL. But this selectivity depended on the detergents used in the reagent.
