Different from a-amino acids, β-amino acids exist in nature only in trace amount, but still, they are a series of important compounds, which lie in their competency to be components of many nature products, the precursors of p-lactams and the building blocks of bioactive compounds. In recent years, with the development of combinatorial chemistry, there is an increasing need for novel β-amino acids that can serve as the building blocks to enrich the chemical library of peptides. Under this circumstance, β-amino acids have being studied more and more extensively.In this paper, 10 β-amino acids are produced, most of them are reported for the first time in our country. Furthermore, to meet the need of the solid-phase synthesis of peptides, protecting groups(Fmoc-) are applied to protect the synthesized amino acids, therefore derivatives are obtained in high level of purity.Rodionov method is applied in this paper, that is, corresponding aldhyde, malonic acid and ammonium acetate are used to synthesize P-amino acids. In the production, Rodionov method is investigated carefully not only in various reaction conditions such as ratio of materials, temperature, time, solvent and so on, but also in reaction mechanism. As a result , the yield of p-p-nitrophenyl-alanine is improved from previous 40% to 60% by the improved operation method and solvent. Moreover, satisfying results have been reached with different separation methods on the base of their physical property.In the last respect, the structures of above mentioned compounds are determined with the aid of 1H-NMR, 13C-NMR and MS, and the purity of N-Fmoc-protect β-amino acids is measured through HPLC.
