The compounds of coumarin have many kinds of biological activity, and are important in pharmaceutical chemistry. The synthesis of new coumarin derivatives is vital in order to find higher biological activity compounds. So we prepared there coumarin derivatives (Scheme 1):scheme 11. The development of coumarins bioactivity was reviewed. Organophosphorus (V) derivatives like phosphonates, phosphinates, find great utility as inhibitors of many proteases and other enzymes in life.2. The phosphorylating methods alcohol were summarized. The Atherton-Todd reaction suits phosphorylation for the phenol hydroxyl because of its simplicity, higher productivity and little side-products et al. So we phosphorylated the coumarins by Atherton-Todd reaction.3. All the coumarins selected here were firstly phosphorylated by Atherton-Todd reaction with higher yields in normal conditions. Twelve derivatives of coumarins were achevied. The phosphorylated coumarins were identified by IR, 1H NMR, 31P NMR, ESI-MS. The method was shown as Scheme 1:scheme 24. Part of the products were cultivated into crystal, and their structures were further proved. At the same time, the datum were accumulated for the further study of the structure and character of coumarin phosphates.5. The reaction activity of the hydroxyls of 6, 7-dihydroxy-4-methylcoumrin wasstudied. We found that the 6-hydroxyl had the higher activity than the 7-hydroxyl in the reaction. The experimental result was further proved by quantum chemistry.
