Activated sludge process is the most popular technology in wastewater biotreatment at present, and microbial community in activated sludge is the critical factor affecting the efficiency of the process in most wastewater treatment plants. Therefore, the study of microbial community in activated sludge has always been being the hot spot of environment science, ecology, environmental engineering, and so on abroad. The research on microorganism community at home, however, is just at the beginning stage, far behind that abroad.By comparing the dispersion efficiency of activated sludge floc by grinding in the mortar and shaking with little glass beads, we find that the grinding method releases 1.72 times more microorganisms than the shaking does, and the suspension by the former is suitable to the flow cytometry (FCM) due to its less impurity.The sample of suspension assayed by FCM shows that the α , β and γ subclass of Proteohacteria hold a relative percentage of 15.09%, 20.50% and 12.00% respectively, which have some difference with the reports abroad. It is possible that there is a distinction of germ species and amounts in different water inhabits.After ultraviolet(UV) light (15W, 20cm) irradiates on the activated sludge for 3 minutes, the sections of microorganisms are assayed by FCM and reveal that the viable rates of α -, β -and γ - Proteobacteria , Pseudomonas ssp., Comamonas spp decrease to 9.10% , 10% , 81.89% , 40.31% and 41.16% respectively. It's proved that UV has much influence on the structure of communities in activated sludge.The implantation of N+( 30KeV) in the flocs has a more notable effect on the structure than the UV: under the dose of 1× 1015ions/cm2, Comamonas spp have extincted and α - Proteobacteria are rarely found; when the dose increases to 1× 1016ions/cm2, besides Comamonas spp., Pseudomonas ssp. and α - Proteobacteria have perished completely.It's the first time at home to apply FCM to analyze the changes of microorganismcommunities mutagenized by low-energy ion beam and UV, so as to give us a useful reference for determining the suitable dosage or other problems when we engage in microbial community breeding by UV and low energy beam.
