Flutriafol Synthesis

Flutriafol is a good fungicide useful in plant growth.Flutriafol was prepared using fluorobenzene and o-fluorobenzoyl chloride as raw materials by Friedel-Crafts acylation, Corey-Chaykovsky reaction and 1,2-Epoxide Opening reaction in basic conditions.At last,flutriafol was synthesized.In the experiment, the high yield can be gained through making optimized condition to the several reactions.In the first Friedel-Crafts acylation,using fluorobenzene as raw material and solvent,the optimized conditions are as follows,reaction time 3h,reaction temperature 84℃ and mole ratio of catalyst and o-fluorobenzoyl chloride 1.2,so 91.9% yield can be gained.In the second sulphonium salt preparation and Corey-Chaykovsky reaction, the optimized conditions are as follows,mole ratio of dimethyl sulphide,methano -l,vitriol,potassium hydroxide and 2,4'-difluorobenzophenone 3.2:1.6:2.4:8:1,reaction temperature 45℃,so 92.5% yield can be gained.In the third 1,2-Epoxide Opening reaction,the optimized conditions are as follows,mole ratio of l-H-l,2,4-triazole and 1-(2-fluorophenyl)-1-(4-fluorophenyl) oxirane 2,reaction temperature 80℃,reaction time 4h,solvent DMSO,so 72.3% yield can be gained.This paper bring innovation in Corey-Chaykovsky reaction,as easy-prepared and active sulphonium salt (CH₃)₃SHSO₄ was used.Furthermore,the configuration of the impu -rity in the third reaction was confirmed and the selectivity was studied simply.
