Ginkgo Biloba is one of characteristics in China, Ginkgo Biloba has absolutely advantage of resources in China. 90 percent of Ginkgo Biloba are located and 70 percent of Ginkgo Biloba Leaves are produced in China. The Ginkgo Biloba Leaves consist of many active components, including flavonoids, terpenes, which are used widely in medicine, food, chemical engineering. Planting area of Ginkgo Biloba become increasingly large, it is famous for the products of Ginkgo Biloba as natured medicines, preparation of Ginkgo Biloba Leaves became one of resources of importation medicines in America. At present, more than 60 factories produce preparation of Ginkgo Biloba Leaves, they need a lot of standardized extracting. Some countries have no Ginkgo Biloba and want to produce preparation of Ginkgo Biloba mainly depend on China. Although EGB output increasingly by year, sale price is on the decline. In this paper, on base of the modern technology of extraction and separation, the process of separation flavonoids and terpenes from Ginkgo Biloba Leaves is developed.There are four fractions in this paper as follows:( 1 ) Determination of three flavonoids aglycones by high performance liquid chromatography is developed. Hydrolysis conditions is determined by response surface methology, the constitutes of flow phase, acidity, flow speed and quantity of injected sample are optimized.The method is accurate, rapid and reliable. At the same time, spectrophotoetric determination of flavonoids is studied, which can be used to determine the content of flavonoids from Ginkgo Biloba Leaves or extraction in ranges of linear line. What's over, a reversed-phase high performance liquid chromatography method is established for simultaneous determination of terpenes, the constitutes of flow phase and flow speed are optimized.(2) The process of extraction, separation of flavonoids is studied in detail. The efficient factors of extracting yield of flavonoids by experiments of the single factor and orthogonal experiments are optimized. The optimum conditions of extraction as follows: concentration of alcohol 70%, extracting temperature 80 °C, extracting time lh, ratio of solid to liquid 1:30, extracting times 1, under this condition, the extracting yield of flavonoids is 70.02%, simultaneously, the extracting total flavonoids from Ginkgo leaves by enzyme hydrolysis was studied,the content of enzyme 0.4mg/mL,time of enzymatic hydrolysis 120min,temperature of enzymatic hydrolysis 55°C,PH value of media 4.5,compared with the traditional extraction of 70% alcohol , extracting yield of the total flavonoids is increased by 6.26%.(3 ) In the process of purification, the macro porous resins as the adsorption are used. Choices of Categories of resins, parameters of static and dynamic adsorption are studied, the purification technology ofextracting Ginkgo Biloba Leaves is established. The results shown that AB-8 has good separation and purification of flavonoids and terpenes, It can be used to prepare Ginkog Biloba Extraction, content of flavonoids is less than 24%.(4) In the process of purification of terpenes, liquid-liquid extraction and silica gel column chromatography are combined in sequence, The efficient factors of extracting yield of terpenes by orthogonal experiments are optimized. Extracting times 4, extracting temperature 25 °C, extracting time 1 Omin, recovery yield 82%;Petroleum ether: ethyl acetate (3:7) as eluting reagent, content of terpenes is 87%.The study will be a new way and idea for Ginkgo Biloba Leaves exploiting.
