Objective Synthesize the (R)-4-[(1s,2s)-1-benzyoxy-2-dibenzylaino-3-(3,5-Difluorophenyl)-propyl]-2,2-dimethyloxazolidine-3- carboxylic acid tert-butyl ester (BDCE) , and optimize this technology on the basis of the experiments.Methods D-Serine and 3,5-difluorobenzaldehyde as starting materials, the intermediate BDCE has been synthesized after ten steps by condensation, alkylation, reduction, oxidation and so on., and the structure has been confirmed by LC-MS,1H-NMR,13C-NMR.Results The samples obtained in each reaction steps all correspond with the chemical structure of the target compound. That proves ,the syntheses of (R)-4-[(1s,2s)-1- benzyoxy-2-dibenzylaino-3-(3,5-difluorophenyl)-propyl]-2,2-dimethyloxazolidine-3- carboxylic acid tert-butyl ester (BDCE) is completed.Conclusion This method synthesizes the (R)-4-[(1s,2s)-1- benzyoxy-2-dibenzylaino-3-(3,5-difluorophenyl)-propyl]-2,2-dimethyloxazolidine-3-carboxylic acid tert-butyl ester (BDCE) .The synthetic route has the characteristic of moderate reaction condition ,simple operation ,cheap available raw material and high yields for each reaction steps, and is applicable in the industrial production. |
