β-carotene is the precursor of vitamin A and has important physiological effects on animals and humans.Comparing with the synthetic compound,naturalβ-carotene is much more popular for its stronger activity and lower poison.Naturalβ-carotene usually can be obtained from plants,algaes,and some waste of factories.In this work the separation ofβ-carotene from the waste of chlorophyll factory were studied.β-carotene was determined by HPLC using YMC-Carotenoid C30column 4.6mm×150mm(3μm).Mixture of methanol and methyl tertiary butyl ether(7:3)was used as elution solvent with flow rate of 0.6ml/min.the wavelength of UV detector was 450nm/205nm.The separation ofβ-carotene from crude chlorophyll factory residue consists of three unit operation including extraction,preparative chromatography and recrystalliztion.During the pretreatment process,the pigment was extracted by ethanol for getting rid of the unsoluble protein and lipids.Then N-hexane/ethanol/water(10:10:1)was selected to extract most foreign matter from excrementum bombycis.Before the raffinate was purified by column chromatography, the performance and separation characteristics relevant toβ-carotene were measured on HZ816,H103,D4006,D4020,D14,X5,S8,AB8,HZ801,DK110resins. It was found that X5 and HZ816 macroporous adsorptive resins offer the largest adsorption abilities among the tested adsorbents and thus were selected as the stationary phase.Using ethanol/ethyl acetate(or ether)as mobile phase,the content ofβ-carotene was 4.19%with a recovery yield of 56.4%.Through crystallization process in the mixture of chloroform and ethanol at -18℃,a final crystalline product was obtained with the purity higher than 80%.
