Pullulan is a major polysaccharide produced by strains of Aureobasidium pullulans. It is transparent, colourless ,tasteless ,odoourless, tenacious,resistant to oil and grease and unaffentde by small thermal variations.It can be produced by fermenting with sugar such as hydrolyzed amylum,sucrose and glucose. The pullulan has the characteristics of water-soluble, no-gelation, no-aging and without any toxicity. Pullulan is one of the few neutral water-soluble microbial polysaccharides that can be produced in large quantities by fermentation.It can be widely used in foodstuff,cosmetic,energy production, pharmaceutical industries, nicotiana tabacum and agriculture.It can be decomposed by certain microorganisms and will not pollute the envioment. It is multifunction material of having the largest economic value and exploitation potentiality.China is the largest producer of MSG.. Glutamate wastewater is a kind of high concentration organic wastewater. With high acidity, high ammonia concentration, high sulphate concentration and high organic pollutant concentration, glutamate wastewater could not be treated successfully with simple traditional method. However, many of which contain nutrients.By this experiment, we analyze ion-exchange wastewater which can be produced in the process of manufacturing monosodium glutamate, and we attempt to use ion-exchange wastewater as the fermentation matrix to produce pullulan. The results in detail were stated as follows:1. MSG liquor in the biological transformation should be carried out in the department. glutamate wastewater from the high-speed centrifuges used centrifuge to remove solids.By adding powdered lime (CaO), the addition of lime can make liquor in SO42+ precipita tion, and then centrifuge to remove sediment.Also in the magnetic stirring under the access to air stripping, the mother liquor in NH3-N to NH3 were in the form of stripping, making liquor pH gradually increased.quicklime amount of 1.5 %(quality and volume ratio), SO42+ removal rates of up to 70 %to 80 %, the wastewater from the pH value of around 1.5 to 6.5. 2. The optimized culture conditions are as follows:initial pH6.5,pullulan cells transferred to 500ml conical flasks containing 150ml of the culture medium; inoculation incubated according to 10% of the inoculum; in a rotary shaker incubator at 200rpm at 28℃for 7 days.As a result, 23.5g/L Crude pullulan is obtained,and the conversion rate is 47% . By shucking off pigment through activated carbon column and then protein removal, the recovery rate of pullulan reaches 65% .3. This section of the system in the cell protein powder, including monosodium glutamate in the cell and waste water fermentation pullulan polysaccharide after the cell in two parts. Production of pullulan polysaccharide cell can be recovered after the amount of 150 g / L, the crude protein content of about 10.6%; MSG wastewater recycling to the cell to 83.5 g / L, the crude protein content of about 7.5 %.
