Some contents of peptide was simply mentioned at the front part of the thesis, including peptide's concept, synthesis's history, kinds methods of synthesis, basic principle, the modern conditions of domestic country and foreign country, methods of analysis and purification, and its research significance. Solid-Phase Synthesis and purification of sleeping peptide and osteogenic growth peptide were studied in this thesis. Through systemically studying the effects of resin type, coupling time, coupling solvents, coupling reagents and cleavage conditions on the yield of sleeping peptide and osteogenic growth peptide during Fmoc solid-phase peptide synthesis, the protocol was optimized. Using DMF as the coupling solvent; TBTU as the coupling reagent; TFA, p-methyl phenol, EDT and H2O (97:0.25:0.25:2.5) as the cleavage solution's confecting condition, under 3 equiv, amino acid at a coupling rate of 3h per ammo acid. The sleeping peptide and osteogenic growth peptide can be synthesized successfully under this condition. The sleeping peptide and osteogenic growth peptide were isolated and purified, and the optimal technology was established: The crude product was purified by gel chromatography fist with the purity of 60%, and was completely purified by reversed-phase high performance liquid chromatography (RP-HPLC) again with the purity of 95%.After that, one of the aims of the thesis is to introduce a piece of amino acids on the base of the original sleeping peptide, hoping to make it passing through the cell membrane easily, and its biological activity was tested. But the result is not good as we expected. The biological activity of osteogenic growth peptide was not tested because of less time. It need test in the future job. Some of the trouble of synthesis was preferred in the thesis, which was analyzed also. Some suggestions were preferred at the end of the thesis.
