Dicyclopentadiene was decomposed into monomer(cyclopentadiene) by the method of vapor-phase thermal decomposition. Then titanocene dichloride was synthesized via the reaction of cyclopentadiene and TiCl4 under the existence of diethylamine. The influence of the reaction, such as the solvents, the mixture ratios of reactants, the amounts of diethylamine, were studied. The yield of titanocene dichloride reaches 75.4%when the molar ratio of diethylamine:TiCl4:cyclopentadiene was 3:1:4,taking dioxane as the solvent, atmospheric refluxing 4h.The IR spectrum,UV-vis spectrum and 1H-NMR were taken to characterize the compound.Due to the poor solublity of titanocene dichloride in water, some hydrophiliuc groups are colvaleted to titanocene molecule. In this study, three compounds, namely bis(cyclopentadienyl) salicylato titanium(Ⅳ) (TSC),Titanocene di(L-methionine) dichloride(TMC) and Titanocene di(L-phenylalanine) dichloride(TPC),which contained hydrophilic groups were synthesized. These compounds were characterized by elemental analysis,IR spectrum,1H-NMR and UV-vis spectrum. The experimental results showed that TSC was an unimolecular bidentate titanocene compound,TMC and TPC were bimolecular unidentate titanocene compound and possessed good water soluble properties. At the same time, after electrochemistry test,TMC and TPC are of good electrochemistry activity and behave quasi-reversible electrode redox reaction in the experimental potential scope.Titanocene di(L-methionine) dichloride(TMC) is a new kind of antitumor metallorganic compound.Choosing deoxyribonucleic acid (DNA) as the research target. The interaction between TMC and DNA was investigated through the methods of electrochemistry,UV-Visible absorption spectrum and viscometry. The CV results showed that the electroactive complex was formed via electrostatic effects at pH 3.0. Moreover,The number of binding ratio n = 4.26 and an affinity constant of Ka =2.74×1016 were obtained. At the same time, the results obtained by UV-Vis spectroscopic and viscometry also indicated that the interacton way between TMC and DNA was electrostatic effects.
