Cucumber mosaic virus (CMV) is a typical single-stranded, plus sense RNA virus, and has an extremely large host range and has a world wide distribution, one of the pathogenicity impaired crops. It has three genomic RNAs. So it is one of RNA virus model strains to investigate pathogenicity mechanism in host.CMV is one of the most important pathogen which cause processing tomato to reduce production or even not to have production, conditional resist virus breeding and agrochemical prevention and cure not only cause environmental pollution but also waste time and strength. But RNAi technology posses superiority which conditional gene study don't have, it can provide a new pathway to study Gene function and Plant gene expression and regulation, it also can depress exogenous gene expression selectively, so RNAi technology show its peculiarity aspect in virus and pest prevention and cure.NS04 CMV RNA1-3 genome sequence was amplified which was isolate from processing tomato by RT-PCR. Using phylogenetic tree analysis, the result showed that, four protein from NS04 genome CMV-1a had a high homology of nucleotide and amino acid to AB179764 from Japan, which is 96.0% and 98.6%; CMV-2a had a high homology of nucleotide and amino acid to DQ41273 from Zhejiang, which is 98.0% and 96.5%; CMV-2b had a high homology of nucleotide and amino acid to S72187, which is 95.5% and 90.1%; CMV-CP had a high homology of nucleotide and amino acid to EF312025 from Zhejiang, which is 93.7% and 98.1%. This result certify CMV variate easily, and CMV could adapt new host ane environment fastly, so strain differentiation of CMV is very complex.Selecting replicative enzyme of RNA2 as target sequence, two section of specificity fragments were synthesized and pBi35SCR2 RNAi eukarya expression vector was constructed successfully. The vector was transformed to Nicotiana tobacum by Agrobacterium tumefaciens mediate method. Obtained transgenic Nicotiana tobacum could be used not only as materials for virus attack test, but also used to lay a foundation for studing processing tomato against Cucumber mosaic virus.
