Objective: (1)Based on six normal varieties of cucumbers and one mutant of virescent,we counted the cell chromosome numbers and studied the karyotype analysis of cucumber which provided a reference on the gene location related the cucumber mutant of virescent.(2) Discussion about the differentially expressed genes between the mutant of virescent and wild-type leaf tissue.Method: (1) we analyzed the results which we prepared chromosomes using cucumber roots by conventional compression method, and then made the microscopical examinations and micrography.(2) The subtractive hybridization library of the virescent mutant cucumber was constructed by SSH.Identification the clones of suppression subtractive hybridization library by restriction enzyme digestion and sequenced the clones whose fragments are between 200~900bp by restriction enzyme digestion. The sequences were blasted in the GenBank Nucleic Acid Database which were removed restriction sites.Result and conclusion:(1)In accordance with the normal and the mutant cucumber karyotype analysis,the results showed that the normal and the cucumber mutant of yellow virescent's chromosomes are both 2n=14, that belong to diploid plant and the basic number of chromosome is 7. Karyotypes belong to 1A and they are symmetrical karyotype.The variance analysis of different of cucumber varieties of chromosomes'length showed that P>0.05.It showed that there was no difference between normal and the cucumber mutant of yellow virescent at the level of chromosomes.(2)We identified a total of 240 positive clones, including 152 forward subtractive library positive clones and 88 reverse subtractive library clones.206 sequences have been sequenced successfully.After blasted analysis of 159 sequences which were transformed to the FASTA format and submitted to the GenBank,at last we got a serials of numbers from GH270133 to GH270291.
