Epigenetics is a descendible gene expession without the variational DNA sequence. DNA methylation is one important part of epigenetics. Aslo, the research is indicated that the level of DNA methylation is a mutative course in fetation; some important course such as character differentiation in fetation. so there is a technique by MSAP(methylation sensitive amplified polymorphism) to analyse the change of methylation sites in chicken fetation. First, apply the technique to fowl fetation to find the variation of significant informations Epigenetics. Then, we can find the data of change direction in chicken DNA methylation and the difference in different tissue and apparatus for the first time. By the process, it shows that there is a ralation bewteen dynamic rule of DNA ethylation in chicken fetation and the develop of apparatus. And And it's the foundation of the effect of economic character on researching the DNA methylation and other epigenetics.Chicken embryo is test materia in the research. Extracted DNA from Gallus domesticus embryo, then detected the level of methylation by MSAP. The result shows that the number of methylation sites are 880, and the sites without double-stranded and single-stranded DNA methylation is 1332, in which the rate of single-stranded methylation (hmCCGG) in outboard is 20.48%, double-stranded DNA(Cm CGG) in side is 19.30%, respectively, and the total of them is 39.78%. And the methylationsite rate is 32.96% in 3 days embryo, 38.48% in 5 days, 39.45% in 12 days, and 47.73% in 15days. After all, the direction of advance with the level of DNA methylation is along with fetation.The tissue and apparatus of 15 days Chicken embryo is test materia in the research. Extracted DNA from Gallus domesticus embryo, then detected the level of methylation by MSAP. The result indicates that the number of methylation sites are 1379, and the sites without double-stranded and single-stranded DNA methylation is 1553, in which the rate of single-stranded methylation (hmCCGG) in outboard is 23.67%, double-stranded DNA (Cm CGG) in side is 23.36%, respectively, and the total of them is 47.03%. And the methylationsite rate is 47.73% in tissue of chest muscle in embryo, 42.74% in heart, 39.45% in lung, 46.64% in kidney, and 52.48% in liver After all, the direction of advance with the level of DNA methylation is along with fetation.In summer, the DNA methylation of chicken embryo is increased by degrees by the time of development and growth: the rate of methylation variety is more from the 12 to 15 days, and not as markedness from 5 to 12 days. On the other hand, the chicken embryo is presented a significant difference between the different tissues. The level of methylation is high in liver and low in lung, the other tissues with examined is adjacence. The time line of methylation in embryo is accord with gain weight, and the change of tissues difference is also accord with the dynamic course of apparatus development. And the result is approved that the fastigium of methylation may in the time of morula, then the process of methylation with tissues difference is always go with the fetation.
