The strain 814 of Marek's disease virus(MD\ gene encoding glyeoprotein B (gB) has been cloned and the entire nucleotide sequence of the gene has been detennined The gene encoding glycoprotein A (gA) of Fe- 126 strain turkey of herpesvirus (HVT) is inserted by lac Z gene of Escherichia coil. The recombinant HVT containing IacZ gene (lacZ-rHVT) stably expressed AC -galactosidase in infected cells and also could be reisolated from chickens, indicating the gA gene of HVT is nonessential to HVT replication. Furthermore, the glycoprotein gB gene of the strain 814 of MDV was integrated into this region by homologous recombination with IacZ-rHVT as the parent virus. The recombinant was isolated by the white-blue plaque selection and proved by dot-blot and PCR detection. Expression of the recombinant was proved by indirect immuno-fluorescent assay.
