An improved protocol for Agrobacterium-mediated transformation of rice is described. We introduced the gene of GUS by LBA4404 (pTOK233) and obtained a large number of transgenic plants, that were fertile and morphologically normal. The procedure permitted the regeneration of transgenic plants from callus in only three months. Casamino acids and proline are included in the medium for callus induction and in the medium for selecting transgenic calli, and all cultures are incubated under light at 30 0C, except during the co-culture period. These modifications enabled me to regenerate transgenic plants in only three months, starting with callus induction from mature seeds of ZH8 18773 and MH63. Using 2/3MS+1/3YEB to dilute the Agrobacterium clone, we got the transformation efficiency of 20.8%. By adding AS+G we improved the efficiency of transformation to 2 6.9%.
