Adult Goat Skin Stem Cell Isolation And Cloning

There are no report on establishment of goat skin stem cell lines untill now. The present experiment aimed to establish an isolation method and culture system to clone skin stem cells in vitro. The identification and characteristics of the skin stem cells were also studied. The results as follows. 1 .Skin stem cells Could obtained through skin explant culture, synchronous culture and epithelial cell secondary culture.The skin explant culture are better than others. It抯 results could be repeated under similar experiment condition. 2.The culture medium (TCM 199 Supplemented with 20% NBS, penicillin, streptomycin and IGF- 1) is good for the skin cells cultured in Vitro. 3. IGF-l could work to improve cloning the skin-like stem cells. The clone were appeared ahead froml to 2 days when the IGF-l was added with 20-35ng/ml. 4. Skin stem cells has the ability of passaging, The present experiment passaged it two or three passages. 5. The clone had different growth performance in different feeder layer. The goat skin fibroblast cell served as feeder lager was better for clone than epithelial cell's. 6. Skin stem cell could stained to be AKP positive. 7. The skin stem cell clone suspension cultured in Vitro has the tendency to forming embiyoid bodies. 8. Being Cultured in conditional medium made from cartilage explant culture. The skin stem cells could be in duced to differentiate into cartilage cell.
