SSR was used to screen a new powdery raidew resistance gene M53 contained in a durum-Aegilops squarrosa synthetic hexaploid wheat, which facilitates the marker-assisted selection and cloning of the new gene.Molecular markers closed linked to some powdery mildew resistance genes, barley yellow resistance gene~Bdv2, and HMW subunit gene~!Dx5 were used to select and pyramid these superior genes in breeding lines and obtained the following results:1. Molecular tagging of a new powdery mildew resistance geneA total of 72 pairs microsatellite primers .which have been located in D-genome, were used to screen for SSR markers in M53, Shan7859, the R and the S pools. About20.8% primer pairs showded polymorphism between resistance parent M53 and susceptible parent shan7859. One SSR primer, Xwmc289, genereted a specific SSR product in the resistant parent and the resistant pool. Linkage analysis in F2 population with primer pair Xwmc289 showed that the genetic distance between the marker Xwmc289 and the PmX gene was 10. 86 + 2. 35cM. the polymorphic band were successfully clonded and sequenced.2. Marker-assisted SelectionSTS-PCR marker for Pm4, SCAR marker for Bdv2, STS marker for 10x5 and SSR marker for PmX were used to screen 4 breeding population, eight plants was found to pyramid these genes.
