Trehalose has A special function on protection of the structures of the proteins and the membranes. In trehalose-producing organisms, it serves as a protectant from stresses , such as desiccation and freezing.In order to obtain the new variety of the Stylosanthes which contains trehalose and has stronger drought-resistance, Researches were given on genetic transformation of Stylosanthes.Establishment of the regeneration system from leaves to calli to plantlets for the Stylosanthes (Reyan No.2). Genetic transformation of the calli were down.by using agrobacterium mediated transformation method. 6 of the transformants were obtained in this experiment. The transformants were confirmed by the PCR and PCR-Southern hybridization analysis.The results indicated that 2 plantlets could amplified a 2.2kb fragment, and 4 of them could not amplified any fragment. The PCR-Southern analysis showed that the 2 of them were also positive. It demonstrated that the Trehalose synthase gene was integrated into Stylosanthes genomes.lt has a good foundation for breeding drought-resistance Stylosanthes.
