In this paper,a stable and optimal regeneration system of regenerated Cotinus coggygria "Purpureus" with the technique of tissue culture was achieved. Axillary buds as explants were cultured on favorable media.Axillary buds as explants were cultured on improved MS media containing NAA 0.1mg/L,6-BA 1.0 mg/L to induce buds to grow and propagate; plantlets were inoculated on reformative White media supplemented with NAA 1.0 mg/L to induce roots. A large number of regeneratd plantlets were obtained quickly by means of this method.Because the process of the induction of roots was affected by some enviromental factors, we discussed how the three enviromental factors (plant hormone, activated charcoal and value of pH of media) to affect the frequency of induction and growth of roots.The results about the effect of plant hormone showed that NAA evidently accelerated induction of roots in vitro. On reformative White media supplemented with NAA 1 .0mg/L,the frequency of rootage raised from 22.2% on basic media to 88.9%. But the concentration of NAA higher than 1 .0mg/L restrained induction and growth of roots. When the concentration of NAA reaching to 2.0mg/L, the frequency of rootage decressed obviously. This manifested that proper concentration of NAA was important for induction of roots, while higher concentration of NAA controled induction of roots.We also studied effect of activated charcoal on induction and growth of roots. We found that activated charcoal weakened explant browing . For induction and growth of roots,activated charcoal didn't restrain the growth of roots and advance tiller, but addition of activated charcoal on media postponded the form of roots,reduced the frequency of rootage,growed thin and not strong,and didn't form tussocky buds at all.On media without activated charoal, the frequency of rootage and time of formation of root respectively were 88.9% and 7 days.Comparatively, on media with different concentration of 0.5% and 1.0% of activated charcoal , the frequency of rootage was just 38.9%,and the formation of roots prolonged to 12 days and 15 days respectively. So we confirmed that activatedcharcoal was not favorable to induct roots.The value of pH as another factor improve to induct root and grow. At pH6.2,the formation of roots was shortened relatively,as well as the roots growed strongly.
