| Sugarcane mosaic virus(SCMV) is a very serious virus disease for sugarcane growth which can cause serious yield lost and quality decline.Through molecular identification,Sorghum mosaic virus,the main disease source for Sugarcane in China has been testified as one strain of Potato virus Y genus.It's demonstrated by bio-information analysis of the obtained HC-Pro(Helper component-proteinase) protein that HC-Pro protein is stable containing several hydrophobic regions but lower hydrophobicity.No transmemebrance domain was found.Based on the analysis of secondary structure prediction,it's showed that HC-Pro protein consists of many helix structures.In the study,the plant expressing vector of UBI-SrMV-HC-Pro was constructed.The breed of FuNong 95-1702 is susceptible to SCMV.One thousand and five hundred calluses of FuNong 95-1702 were transformed with two vectors by bombardment, UBI-SrMV-HC-Pro containing SrMV-HC-Pro gene and another plant expressing vector-pmi containing pZMLR14,one mark gene for screening.After selection on the medium containing mannose and the following subculturing,differentiating and root inducing,fifty-five transgenic sugarcane individual plants were obtained.The test of pmi gene by Color-reaction of chlorphenolred showed that during the fifty-five transgenic plants,there were twenty-six plants which were positive.PCR analysis showed that 12 individual plants contained the HC-Pro gene during which 10 plants were positive in dot blot hybridization and RT-PCR reaction.But for the 10 positive transgenic plants testified by PCR analysis,the number of positive plants in the color-reaction of chlorphenolred is only nine.The above result showed that one transgenic plant containing HC-Pro gene doesn't contain the mark gene-pmi.Through the study of field survey of 10 transgenic plants,it's found that all the transgenic plants with HC-Pro gene were infected with SrMV.HC-Pro gene may take important plays in the arising of sugarcane mosaic disease. |
